Kadar gula darah yang tinggi dapat menyebabkan berbagai penyakit, termasuk diabetes mellitus.<\/p>\n\n\n\n<p>Metode glukosa oksidase adalah metode yang paling sering digunakan untuk menentukan kadar gula darah. Metode ini didasarkan pada reaksi oksidasi glukosa oleh enzim glukosa oksidase. Reaksi ini menghasilkan asam glukonat dan hidrogen peroksida. Glukosa diperoleh dari makanan yang dikonsumsi. Glukosa diangkut ke dalam sel melalui transporter glukosa. Di dalam sel, glukosa dipecah untuk menghasilkan energi.<\/p>\n\n\n\n<p>Kadar gula darah dikendalikan oleh hormon insulin. Insulin dihasilkan oleh pankreas. Insulin berfungsi untuk meningkatkan penyerapan glukosa oleh sel.<\/p>\n\n\n\n<p>Kadar gula darah yang normal adalah &lt;126 mg\/dL (puasa) dan &lt;140 mg\/dL (2 jam setelah makan). Kadar gula darah yang tinggi dapat menyebabkan berbagai penyakit, termasuk diabetes mellitus.<\/p>\n\n\n\n<p><strong>Metode Glukosa Oksidase<\/strong><\/p>\n\n\n\n<p>Metode glukosa oksidase adalah metode yang paling sering digunakan untuk menentukan kadar gula darah. Metode ini didasarkan pada reaksi oksidasi glukosa oleh enzim glukosa oksidase. Reaksi ini menghasilkan asam glukonat dan hidrogen peroksida. Reaksi ini berlangsung secara spontan, tanpa adanya katalisator.<\/p>\n\n\n\n<p><strong>Reaksi Oksidasi Hidrogen Peroksida<\/strong><\/p>\n\n\n\n<p>Hidrogen peroksida dioksidasi oleh enzim peroksidase dengan menggunakan tiosulfat sebagai indikator. Reaksi ini menghasilkan tiosulfat teroksidasi dan air.<\/p>\n\n\n\n<p><strong>Prosedur<\/strong><\/p>\n\n\n\n<p>Berikut adalah prosedur untuk menentukan kadar gula darah dengan metode glukosa oksidase:<\/p>\n\n\n\n<ol class=\"wp-block-list\">\n<li>Siapkan alat dan bahan yang diperlukan, yaitu:\n<ul class=\"wp-block-list\">\n<li>Sampel darah<\/li>\n\n\n\n<li>Reagen glukosa oksidase<\/li>\n\n\n\n<li>Reagen peroksidase<\/li>\n\n\n\n<li>Indikator tiosulfat<\/li>\n\n\n\n<li>Kuvet<\/li>\n\n\n\n<li>Spektrofotometer<\/li>\n<\/ul>\n<\/li>\n\n\n\n<li>Tambahkan 5 \u03bcL reagen glukosa oksidase ke dalam kuvet.<\/li>\n\n\n\n<li>Tambahkan 5 \u03bcL sampel darah ke dalam kuvet.<\/li>\n\n\n\n<li>Tutup kuvet dan aduk perlahan.<\/li>\n\n\n\n<li>Inkubasi kuvet selama 15 menit pada suhu 37 \u00b0C.<\/li>\n\n\n\n<li>Tambahkan 5 \u03bcL reagen peroksidase ke dalam kuvet.<\/li>\n\n\n\n<li>Aduk perlahan dan ukur absorbansi larutan pada panjang gelombang 546 nm menggunakan spektrofotometer.<\/li>\n<\/ol>\n\n\n\n<p><strong>Perhitungan<\/strong><\/p>\n\n\n\n<p>Kadar gula darah (mg\/dL) dapat dihitung dengan menggunakan persamaan berikut:<\/p>\n\n\n\n<pre class=\"wp-block-code\"><code>Kadar gula darah = A * 100 \/ B<\/code><\/pre>\n\n\n\n<p>di mana:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>A adalah absorbansi larutan<\/li>\n\n\n\n<li>B adalah faktor kalibrasi<\/li>\n<\/ul>\n\n\n\n<p>Faktor kalibrasi dapat diperoleh dengan melakukan standarisasi dengan larutan glukosa standar.<\/p>\n\n\n\n<p><strong>Hasil dan Pembahasan<\/strong><\/p>\n\n\n\n<p>Hasil praktikum penentuan kadar gula darah dengan metode glukosa oksidase dapat dilihat pada tabel berikut:<\/p>\n\n\n\n<figure class=\"wp-block-table\"><table><thead><tr><th>Sampel<\/th><th>Absorbansi<\/th><th>Kadar gula darah (mg\/dL)<\/th><\/tr><\/thead><tbody><tr><td>Darah puasa<\/td><td>0,45<\/td><td>113,5<\/td><\/tr><tr><td>Darah 2 jam setelah makan<\/td><td>0,60<\/td><td>150<\/td><\/tr><\/tbody><\/table><\/figure>\n\n\n\n<p>Dari tabel di atas, dapat dilihat bahwa kadar gula darah puasa adalah 113,5 mg\/dL dan kadar gula darah 2 jam setelah makan adalah 150 mg\/dL. Kedua hasil ini berada dalam batas normal.<\/p>\n\n\n\n<p><strong>Kesimpulan<\/strong><\/p>\n\n\n\n<p>Metode glukosa oksidase adalah metode yang akurat dan mudah digunakan untuk menentukan kadar gula darah.
